TOBI Alum publishes study comparing cellular and cytokine characteristics of PRP and BMC from two different preparation systems
This paper by Dr. Cassano and colleagues at Cornell University College of Veterinary Medicine & Hospital for Special Surgery (HSS) describes the results of a study designed to compare the cellular and cytokine characteristics of platelet rich plasma (PRP) and bone marrow concentrate (BMC) from two different preparation systems.
Each in 29 patients undergoing foot & ankle procedures at HSS had blood drawn to derive PRP using the Magellan®, (Arteriocyte) System, and had enough bone marrow aspirate to derive BMC using both the above Magellan system, as well as the SmartPrep® 2 (Harvest) system. The samples were analyzed using cytology, flow cytometry and cytokine assay kits.
Comparing Arteriocyte PRP vs BMC, it was found that the BMC had markedly increased leukocytes: Neutrophils up by 19x, monocytes by 11x, and lymphocytes by 7x.
Although there were differences in the nucleated cells between the Arteriorcyte and Harvest system, both had equal colony forming unit assays. Many cytokines, such as TGF-β1, PDGF, Il-1β, IL-8, and IL-1ra were present in elevated concentrations in the BMC from both systems.
According to the authors, “the most important finding of this study is the discovery that bone marrow-derived biologics contain clinically relevant concentrations of IL-1ra.” This Il-1 inhibitor is one particular component of autologous conditioned serum (ACS) thought to underpin its clinical effectiveness. Because the creation of ACS is restricted in parts of the United States, it is helpful for clinicians to know that BMC can be a potential source of IL-1ra.
